Chasing a stiff target of 227, Du Plessis was well supported by Glenn Maxwell, who hit a typically belligerent 76 off 36 deliveries.
The chase turned when the pair were dismissed, both falling to excellent catches from wicketkeeper MS Dhoni in the space of two overs.
But, needing 19 off the final over, they fell just short to end the inning on 218-8 in their 20 overs.
New Zealand international Devon Conway's spectacular 83 off 45 and Shivam Dube's 52 helped Chennai post an impressive 226-6 inthe first innings.
The victory moved captain Dhoni's men into third position in the table with three wins from five matches while RCB have won twice in the same number of games.
