Rickelton beat the previous best score by a home player at the venue, which was 228 by Herschelle Gibbs also against Pakistan in 2003.
Rickelton's heroics together with Kyle Verreynne's hundred and Marco Jansen's quick-fire fifty helped the Proteas post a mammoth 615 all out before the visitors were reduced to 64-3 at the close of play.
The hosts resumed the second morning on 316-4 with Rickelton on 176 and David Bedingham on 4 with the latter falling in the fourth over of the morning, edging behind to seamer Mohammad Abbas.
Verreynne then joined Rickelton at the crease and the left-handed batter went to his double-century off 266 deliveries as the pair upped the tempo to score 113 runs in 27 overs in the first session, taking the total at lunch to 429-5.
It was the first double-century by a Proteas batter since Hashim Amla's 201 against England in 2016, which also happened in Cape Town.
The duo then continued to dominate what became a tired Pakistan bowling attack and wicketkeeper Verreynne brought up his fourth Test hundred off 144 balls that included nine fours and four sixes.
After a superb 148-run partnership, the gloveman then lost his wicket to spinner Salman Agha which brought Jansen to the crease and lanky all-rounder piled on the agony for the Pakistan bowlers.
Rickelton was out going for a pull shot having batted a total of 607 minutes and 343 balls with the opener leaving Newlands to a standing ovation from the sold-out crowd.
Jansen went full tilt with the bat as the tired Pakistan fielders scurried around the Newlands outfield, smashing 62 from 48 deliveries as the Proteas went past the 600-run mark.
The hosts then had 21 overs with the ball in the evening session and Kagiso Rbaada once again led the way as picked up 2-9 in six overs, taking scalps of Shan Massod and Saud Shakeel.
Jansen capped off a decent personal day when he bowled Kamran Ghulam with Babar Azam (31 not out) and Mohammad Rizwan (9 not out) still at the crease looking to keep Pakistan afloat.
