The Proteas suffered a mammoth 143-run defeat in their first match on Sunday.
De Klerk, however, believes they can show improvement at the M. Chinnaswamy Stadium in Bengaluru and potentially level the the three-match series on Wednesday.
"I think we're still pretty confident. I think we've had a good couple of meetings, and we know exactly what we need to do," de Klerk told the media in an online briefing.
"It was an unfortunate result the other day, all credit has to go to India for the way they played and put us under pressure, but [this] is a new game and we start afresh, ready for whatever challenge gets thrown at us.
"We are hoping to put up a better performance than we did the other day."
India total of 265/8 proved too big of an ask for the South Africans, who could only muster up 122 all-out in 37.4 overs, with de Klerk, who sat out the first game due to illness, noting they have addressed their shortcomings from Sunday and aim to fix them on Wednesday.
"We didn't get enough runs on the board with the bat, [but] we felt like we did a pretty good job with the ball, restricting them to 260," she added.
"I just think their spinners bowled really well, and we just couldn't get going, so I would say it was just a poor batting performance right throughout.
"Losing three wickets in the powerplay was always going to make it really hard to chase a score of 270, and I think they just put a lot of pressure on the middle order as well.
"We've touched on a lot of things, and hopefully we can better in [today's] game."
