Alcaraz saw off his fellow Spaniard Alejandro Davidovich Fokina in straight sets, 7-6 (7-5) 6-4, although he found the going quite difficult at times.
"It was really tough for me," Alcaraz said after the match. "I started playing pretty well, serving for the first set. I was not able to win [that game]... I was going really fast, a lot of things on my mind. [After the first set] I was talking to myself, positive things, trying to play long rallies, trying to [be there] through the points, trying to feel more comfortable with myself."
Alcaraz moves on to face Great Britain's Dan Evans, who booked his place in the semi-finals with a comeback victory over Argentina's Francisco Cerundolo.
Evans lost the first set, but he responded in high-class fashion to secure a 2-6 7-5 6-3 triumph.
And the other semi-final sees Tsitsipas meet Lorenzo Musetti, who had a walkover in his quarter-final after Jannik Sinner was forced to withdraw injured.
Tsitsipas, meanwhile, wasted little time in dispatching Alex Minaur 6-4 6-2.
