Osorio replaces Jesualdo Ferreira after the Portuguese coach was sacked for the second time this season on 22 March following a poor run of results.
Mansour revealed that Colombian Osorio was one of three candidates to take over the reins at the Cairo International Stadium, with German tactician Heiko Herrlich dropping out of the picture last week after talks broke down
The Zamalek president told the club's TV channel: "Osorio has a strong CV. He guided Mexico in the World Cup and managed to beat Germany.
"He also took the charge of Paraguay national team and won 17 titles in his country with different clubs.
"We hire the coach according to his reputation, but finally he could be lucky or not.
"Osorio was among a final three-man shortlist selected by the club's technical committee with another German and Spanish coach."
The 61-year-old Osorio, who has been out of a job since departing Colombian side America de Cali in March last year, previously managed Mexico and Paraguay.
