Salem Gabre finished in second place in 31:53, with Glenrose Xaba becoming the first South African to cross the Finish Line in third place in 32:12.
Nare broke the record of 31:53 set by Namibian Helalia Johannes, improving her time of 32:11 at the same venue last year.
Speaking through an interpreter, Nare wanted to run sub-31 minutes.
"I'm very happy about the outcome. Even though to be honest I needed someone to help me push myself so that I could run sub-31. That was my plan but all in all, it was a good race," she said.
After missing the first leg in Cape Town due to her participation in the London Marathon, where she secured an eighth-place finish last month, Nare showed why she is the two-time Grand Prix champion.
She described South Africa as her second home, confirming that she would return to participate in the six-race series.
"To be honest this Grand Prix feels like home, so more than the winning, I enjoyed the fact that I'm surrounded by people that I love, that I admire and I like the time that I spend here. So, the fact that I'm taking part is good enough for me more than the winning. Even if I don't win that's not an issue, I just wanna be part of this family."
