The win helped Matsatsantsa move up to the second spot on the league standings, overtaking Orlando Pirates who have a game in hand.
Next, SuperSport will host Stellenbosch FC on May 2. Despite extending their winless run to five matches across all competitions, Usuthu remained 12th on the standings and will look to bounce back when they host Sekhukhune United on April 29.
SuperSport looked threatening from the beginning with their wing play, and Thapelo Maseko scored the first goal in the 15th minute.
In the 62nd minute, Ghampani Lungu scored the second goal for SuperSport.
Bonginkosi Ntuli netted a header for AmaZulu in the 73rd minute, but they were unable to secure an equaliser, and SuperSport won 2-1.
