The Motsepe Foundation Championship side, Spurs, made life difficult for the top-flight hosts, forcing SuperSport to come from behind to claim the win.
The Urban Warriors took the lead through Therlo Moosa, but SuperSport responded with an equalizer from Ime Okon before the end of the first half. Neither team managed to break the deadlock in the second half or most of extra time, until Lebohang Maboe's superb strike sealed the win and ensured the game wouldn't be decided by penalties.
Spurs made an impressive start, taking the lead in the fifth minute with a brilliant solo goal from Moosa. He surged down the right wing, cut inside, and fired a left-footed shot past SuperSport goalkeeper Ricardo Goss at his near post to make it 1-0.
SuperSport gradually gained control of the match and piled on pressure around the 20-minute mark. Spurs' goalkeeper, Neil Boshoff, was forced into two brilliant saves: one to deny Aphiwe Baliti's free-kick and another to stop Nyiko Mobbie's close-range header from a corner.
However, Boshoff could do nothing to prevent Okon from equalizing in the 36th minute. After Spurs failed to clear a free-kick delivery into their penalty area, Okon seized the chance to volley the ball home from close range, making it 1-1.
The second half saw SuperSport continue to dominate. In the 56th minute, Baliti delivered a perfect cross from the left for Samir Nurkovic, but the striker's header went wide of the far post.
United made a double substitution just after the hour mark, introducing Terrence Dzvukamanja and Oratilwe Moloisane, while Spurs brought on experienced Nazeer Allie midway through the second half.
In the 72nd minute, SuperSport again looked dangerous, with Mobbie testing Boshoff with a long-range shot after sustained pressure. The keeper came up with another crucial save to keep the match level.
Boshoff was again called into action in the final minute of regular time, rushing off his line to block Moloisane's effort and send the game into extra time.
Late in the first half of extra time, SuperSport came agonizingly close to taking the lead when Dzvukamanja's strike hit the foot of the post after a scramble in the Spurs penalty area.
At the other end, Moosa had a chance to put Spurs back ahead after a great pass from Ebrahim Seedat, but his shot went wide under pressure from Pogiso Sanoka.
With penalties looming, SuperSport struck in the penultimate minute of extra time. Moloisane raced down the left wing and cut the ball back to Maboe, who rifled home a fantastic shot from the edge of the penalty area to secure a 2-1 victory and a place in the quarterfinals for the Premiership side.
