Martinez and Mazraoui had been under evaluation after both left the field during the second half of Saturday's 3-0 Premier League victory at Southampton. De Ligt also had to be substituted in that game due to cramp.
At a press conference on Monday, manager Erik ten Hag confirmed the availability of the trio, stating: "They are all available, they are good."
Ten Hag also provided updates on Rasmus Hojlund, Mason Mount, and Victor Lindelof, who remain unavailable for the Barnsley fixture as they continue their recovery from injuries.
However, he did not exclude them from the squad for Saturday's league match against Crystal Palace, noting: "We will see how the progress will continue."
Regarding Luke Shaw, who is recovering from a calf injury, Ten Hag mentioned that Shaw would not be part of the cup match.
He commented on Shaw's progress: "He is progressing well. Of course we have a plan in our head when he can be ready. But you're always dependent on how the progress will go and you can't make a suggestion, because there are so many factors why a rehab plan can accelerate or slow down."
