Proteas batter Miller scored his runs off just 18 balls and together with Vijay Shankar (51 not out off 24 balls) helped Gujarat chase down a target of 180 with more than two overs to spare.
India opener Shubman Gill had given Gujarat a solid foundation at the top of the order with 49 off 35 balls but they slipped to 93-3 in the 12th over.
The home team had earlier reached a total of 179-7 in their 20 overs thanks mainly to a superb 81 off 39 deliveries from Afghanistan batter Rahmanullah Gurbaz.
Ireland seamer Josh Little and Afghanistan spinner Noor Ahmed were outstanding with the ball for Gujarat as they captured two wickets apiece.
Former West Indies allrounder Andre Russell gave the Kolkata innings same impetus late on with a swash-buckling 34 from 19 balls that included three sixes.
