The South Africa white-ball team have not played together since a T20 series at home against West Indies concluded on 28 March.
While some players in the Proteas squad have played in T20 leagues around the world during international downtime, Duminy and the coaching staff have had plenty of camps where they have honed some players' skills ahead of the 50-over World Cup in India later this year.
Duminy said: "There is a sense of excitement and willingness to see the work we have put in over the past few months come to fruition.
"The conversations we have had and our skills are going to be tested under pressure. What a great opportunity we have now, especially leading up to a World Cup and to be playing against one of the best teams in the world."
Australia are the current T20 World Cup holders and have a long history of fierce battles against the home side with Duminy experiencing some of those as a player.
The former Proteas all-rounder said while the T20 series and the five ODI matches that will follow is a good opportunity see what thet learned, they would also like to "throw the first punch" against the tourists.
"It is home ground advantage against a unique team, but it is about understanding the things we spoke about and how we implement them in the game," Duminy added.
"All of us are excited because it has been close to five months since we played a game, which is unique for South African cricket. But, importantly, we don't want to find ourselves trying to catch up in a series.
"We want to throw the first punch and identify with the style of play we want to achieve."
