The Citizens have only won three out of 10 league matches this season and currently sit 12th in the standings having only accumulated 11 points.
Tinkler's final match in charge was on Sunday when he saw his charges go down to a 1-0 defeat at home against Amazulu at the DHL Stadium.
A statement from the club said: "Cape Town City and head coach Eric Tinkler have mutually agreed to part ways. The club expresses gratitude towards Eric for the last three seasons where he presided over 2nd, 4th and 5th place finishes.
"We wish Eric the very best for the future. The club will announce a new interim coach as we build up to an exciting new project for the 25/26 season and beyond."
Tinkler has struggled to get City to score goals this campaign with the Citizens only finding the back of the net six times in their 10 league matches with only bottom clubs Richards Bay and Magesi having scored less with four apiece.
Cape Town City will appoint an interim head coach ahead of their clash against Kaizer Chiefs on Sunday while they take host Orlando Pirates just four days later.
