Mfundo Vilakazi came off the bench to be the match-winner for Amakhosi with his goal in the 86th minute at the Moses Mabhida Stadium, just two minutes after Vuyo Letlapa was dismissed for a rash challenge.
Amakhosi boss Nasreddine Nabi had made four changes for this tie following their recent loss to Golden Arrows, with Samkelo Zwane, Pule Mmodi, George Matlou and Rushwin Dortly all recalled to the squad.
A fresh Chiefs looked lively early on and Dortley came close to handing his side the lead with a header from a corner that was well saved by Badra Ali Sangare in the Sekhukhune goal.
Sangare then denied Wandile Dube from point-blank range as Chiefs continued to look threatening.
Andy Boyeli was lively for the away side but he was unable to add to his recent goalscoring heroics as his near post shot was collected by Chiefs stopper Bruce Bvuma.
Sangare was kept busy throughout the first half and made a strong save at the end of the opening period to keep out a Tebogo Potsane shot, after a superb through ball from Matlou.
Yusuf Maart came on for Matlou in the second half, and was also agonisingly close to an opener, only to see his shot flash past a post.
Fellow substitute Mfundo Vilakazi, on for Potsane, also tried to make his mark, but he too was denied by Sangare, after a mazy run to enter the box.
But Letlapa's attempts to stop Vilakai saw him dismissed, and Chiefs soon made the most of the one man advantage as Vilakazi was able to find some extra space and finish with a well placed shot past a despairing Sangare in the Sekhukhune goal.
The result saw Chiefs move up to fifth place on the league standings with 21 points from 14 games. Babina Noko are in fourth with 23 points from 13 games.
