The victory continues an impressive campaign for Stellies who have already won the Carling Knockout trophy while they also sit second in the league standings.
Steve Barker's charges dominated their visitors from start to finish at a blustery venue and took the lead as early as the seventh minute when Anicet Oura found the back of the net after a scramble in the box.
The hosts should have added to their tally but were guilty of squandering chances in front of goal before the halftime interval.
Stellenbosch showed their ruthlessness after the interval though and scored another three goals in the first 15 minutes of the second period to kill off the tie.
Devin Titus made it 2-0 after a sweeping move that saw the winger finish from a tight angle while Andre de Jong added another goal not long after.
In their need to get back into the cup tie, the visitors were leaving wide open spaces at the back and the home side took full advantage.
Iqraam Rayners completed the scoring on the hour mark with a smart finish that left SuperSport goalkeeper Ricardo Goss with no chance of saving.
Stellies join Mamelodi Sundowns in the last four of the Nedbank Cup with Orlando Pirates still to play Amazulu and TS Galaxy set to take on Chippa in the remaining quarterfinals.
