The 24-year-old is understood to have been fatally shot during attempt by criminals to take his vehicle while at a petrol station in Roodepoort.
South African Police Service spokesperson Mavela Masondo revealed that two armed suspects approached Fleurs and instructed him to get out of his car before shooting him.
Despite being rushed to hospital, he was pronounced dead upon arrival.
One of the suspects is said to have driven off in the vehicle, while no arrests have been made yet.
Fleurs, born in Cape Town, was a product of the Ubuntu Football Academy and previously played for SuperSport United.
He joined Chiefs on a free transfer after leaving SuperSport at the end of last season, but was yet to make his debut for the Soweto giants.
"It is with a great deal of sadness that we announce that Kaizer Chiefs player, Luke Fleurs tragically lost his life last night during a hijacking incident in Johannesburg," Chiefs said in a statement.
"Our thoughts and prayers are with his family and friends at this difficult time.
"The SAPS are handling the matter and further details will be communicated in due course.
"May his dear soul rest in peace."
