Usuthu will be hoping to move away from the bottom half of the table, while Sundowns want to retain their spot at the summit.
Mkhulise said AmaZulu will not be a walk in the park.
''We are expecting a difficult game because they want points and they are playing at home. They have very good players, they have been playing well but sometimes they don't get good results but it is a good team,'' said Mkhulise.
AmaZulu recently signed midfielder Bongani Zungu on a free transfer after leaving Sundowns six months ago.
Mkhulise believes Usuthu have done well to beef up with the former Glasgow star.
''Bongani is a very good player, he will add value to AmaZulu. We need to be prepared because we do not know if they might decide to play him. We know what he can do on a good day,'' added Mkhulise.
With the fixture right in the middle of the holidays, Mkhulise asked supporters to come in their numbers to Moses Mabhida.
Mkhulise also sent a strong message on safety during this time of the year.
