Atleti, who were 3-1 winners against city rivals Real Madrid last weekend, took the lead through Antoine Griezmann's 20th-minute opener from a tight angle.
Tempers flared in the second half when David Garcia had a goal ruled out for a foul on Atleti midfielder Axel Witsel, which led to the dismissal of Osasuna boss Jagoba Arrasate in the 78th minute.
Rodrigo Riquelme calmly slotted home three minutes later to double Diego Simeone's side's advantage before a flashpoint between Alvaro Morata and Chimy Avila saw both teams reduced to 10 men.
The two clashed in the 84th minute, with Morata getting booked for an aggressive shoulder charge on the Argentinian as the ball went out of play. From the resultant throw-in, Morata held the ball by the corner flag where he caught Iker Munoz with an elbow and as both players fell to the ground, Avila stamped on the striker's leg.
Avila's horror tackle resulted in a straight red card, while Morata was shown a second yellow. Atleti held on to sit six points behind surprise leaders Girona with a game in hand.
Celta Vigo climbed out of the relegation zone, albeit on goals scored, following a 1-1 draw against Alaves.
Rafa Marin's own goal put Celta ahead in the 35th minute, but the home side were reduced to 10 men when midfielder Luca de la Torre was sent off for a second bookable offence. It proved costly for Rafa Benitez's side as Samu equalised for Alaves with 17 minutes left.
Granada remain in the bottom three despite a 1-1 draw at home to Real Betis.
Betis winger Assane Diao looked to have inflicted another defeat on Granada, who had lost five of their opening six league matches, but former Reading striker Lucas Boye earned a point for the hosts.
