The Brazilians face their neighbours on Saturday at the UJ Soccer Stadium in Soweto.
Victory for the defending champions will leave them just a point shy of retaining their trophy, with two games remaining.
Mbane says even though they won their last meeting, clashes against Tuks are always tricky and past results mean nothing.
"The Tshwane Derby is always tricky, yes we won the last one but that result will not count tomorrow. It's gonna be a tough one and it's gonna need us as Sundowns Ladies to switch on because these three points are very important because if we get them then we focus on the next game. So, this game is very important for us and we need to get the three points," Mbane said.
The reigning league champions have won 13 matches in a row and Mbane says it was not an easy feat to achieve.
"Also, 13 wins is not easy but Sundowns is the home of champions so we are expected to win. It was not easy but our goal is to win 13 more games in a row," she added.
The Banyana Banyana defender admits, however, that they did not have a good start to the season but what is key is that they managed to collect when needed.
"Yes Sundowns didn't start the season well but the most important thing is accumulating points and then being on top of the log table. I think that's the most important thing and other teams are improving which is very good for the competition in the league."
